So today, we will learn that HER2 overexpression is a distinct and actionable alteration in non-small cell lung cancer, and standardized ICH-based biomarker testing is essential to identify patients who may benefit from HER2-targeted ADC therapy.
1:19
HER2-targeted ADC therapy has demonstrated clinically meaningful and durable responses in patients with HER2 overexpressing non-small cell lung cancer, representing a notable shift in the treatment of this patient population.
1:36
And a proactive, multidisciplinary approach to monitoring and managing AEs, particularly ILD, combined with structured patient education is critical in maximizing the benefits of HER2-targeted ADC therapy in HER2-overexpressing non-small cell lung cancer.
1:56
And then without further ado, I would ask Dr. Horinuchi, who is director of the Center for Education and Professional Career Development and assistant chief of the Department of Thoracic Oncology of the National Cancer Center Hospital in Tokyo, Japan, to come to the stage.

Dr. Horinuchi.

What do we have?
2:47
So, uh, Egbert, uh, what are the most relevant studies, uh, currently being conducted in patients with non-small cell lung cancer, uh, with HER2 mutations?
2:59
So I think, um, we have-- we already have seen the results of, um, these novel small molecule tyrosine kinase inhibitors, like son- son德尼尼b and sifu德尼尼b, in patients who have been... already been treated, pretreated with chemotherapy plus or minus immunotherapy.
3:20
And the same applies to this, um, uh, ADC called trastuzumab deruxtecan, which also has had, as David Planchard obviously has told you, um, has significant activity in patients with either a HER2 mutant or HER2 overexpressing non-small cell lung cancer that has been treat-- pretreated with chemotherapy plus or minus immunotherapy.
3:46
So the most important question now is: Do these agents have similar or even superior activity against, uh, HER2 mutant non-small cell lung cancer in the metastatic setting in the first-line treatment? And, um, a couple of studies are, um, already recruiting patients for some time and are expected to be, uh, to report, um, very soon.
4:13
I think, um, there was a recent press release that, uh, told us that trastuzumab deruxtecan may have positive results in the first-line setting as compared to, uh, standard of care chemotherapy plus or minus immunotherapy, which will be presented, uh, what I hear, at the next, uh, World Conference of Lung Cancer in Seoul.
4:38
So very excited that that study, uh, is going to, uh, report.
4:43
And I'm hopeful that that study will be positive, as we know from previous experience that chemotherapy plus or minus immunotherapy in this patient population is not very effective in terms of either response rate or overall survival.

12:25
And coming back to, uh, patients with, uh, HER2 mutations, you know, uh, what would be the best, uh, treatment sequence for these, uh, patients with HER2 mutations, ADCs followed by TKIs or TKIs followed by ADCs do you think?
