So LGTX 101 is a selectivity-directed T-cell engager targeting nectin-4 as well as CD3.

Angus SinclairGUEST
And nectin-4 is a target that has been clinically validated, um, and there are approved drugs, uh, antibody drug conjugates, uh, to, uh, nectin-4 that have been successful in the clinic, but really have had challenges in terms of, uh, safety and tolerability and durability of responses in patients that have nectin-4 positive urothelial carcinoma.

Angus SinclairGUEST
So we came in with a challenge to try and create, um, a, um, different mechanism of action to target patients that may have relapsed or are refractory to, uh, this, uh, antibody drug conjugate, but really have a highly effe-efficacious molecule, but really have no safety concerns in terms of targeting the healthy cells that express low levels of nectin-4, which are primarily the keratinocytes.

Angus SinclairGUEST
And this has been observed with that, um, approved antibody drug conjugate, uh, padcev, that there is skin toxicity that's associated with that low level of nectin-4.

Angus SinclairGUEST
So LGTX 101 really has been designed and selected using our machine learning algorithms and high throughput automation and cell-based assays to have a very high degree of selectivity.

A- and what do you think it would take to make multi-specific design feel more like an engineering discipline rather than an art?
