I am joined today by my colleague, Dr. Marcelo Kugelmas, who serves as the Director of Hepatology and Research at South Denver GI and is also a board member for the Chronic Liver Disease Foundation.

Hi, Christina.

Arsal, can you briefly review for us the Phase III GLSEN study, including efficacy and safety?

Happy to do that.

So GLSEN was recently published earlier this year in The Lancet Gastroenterology and Hepatology.

It was a randomized, double-blinded, placebo-controlled, multi-center, multi-part Phase III trial.

First results from the CAP3 multicenter randomized double-blind non-inferiority trial.

which was published in the Lancet Gastroenterology and Hepatology.

CAP3 addresses a very important clinical question that has remained central to Lynch syndrome prevention since the publication of CAP2, whether a lower daily aspirin dose can retain the cancer preventative benefits seen with a higher 600 milligram dose while reducing adverse effects and making long-term prescribing more acceptable in routine

care.

So I completely agree.

Professor Sir John Byrne, thank you very much for joining us and for sharing the findings, interpretation, and clinical implications of CAP3.

Trying to summarize the importance of the genetic evaluation of children with BFEC, I will present you with some of the most relevant results from the NAPID study.

This study was published in Hepatology in 2021.

And it contains the data of about 130 patients with FIC1.

As you can see, the attempt here was to try to classify the patients according to the genetic background, to the genetic mutations, in three different groups, FIC1A, missense formation, FIC1B, one predicted protein truncating mutation, FIC1C, two predicted protein truncating mutations.

But as well, it was important that some predictors of native liver survival at 10 years were clearly identified, such as serum bile acid at presentation higher, 194 micromole per liter, and total serum bilirubin at presentation higher than 72 micromole per liter.

A similar study was published in Journal of Hepatology in 2020 on more than 250 patients with BSEP.

Here, three groups were created, where BSEP1 were patients with two common European mutations that were associated with some residual BSEP protein functionality.
